It sounds like you're using Vista or Windows 7 and you have the game installed to the Program Files or Program Files (x86) directory.

The best thing to do is to move the game directory somewhere else, like C:\EQ which will allow you to change the files in that directory easily as well as avoid any file virtualization issues in the future.

If you don't do that, you will either need to run Notepad as administrator to be able to save the file, or you'll have to copy the file somewhere else, like your desktop, change it, and copy it back.
